p38 MAPK는 비인산화 상태에서는 비활성화되어 있으며, Thr-Gly-Tyr 모티프의 이중 인산화에 의해 빠르게 활성화됩니다.
네 가지 isoform이 존재하며, 조직 발현 및 상위/하위 조절 인자에 대한 친화성이 다릅니다.
세포가 TNF, IL-1, 열충격 등 자극을 받을 때, MAPK kinase-3/6이 Thr180 및 Tyr182를 인산화하여 p38 MAPK를 활성화합니다.
활성화된 Phospho-p38 MAPK는 ATF-2, CHOP-1, MEF-2 등의 전사 인자를 인산화하여 활성화합니다.
