열 매개 항원 회복(heat-mediated antigen retrieval)을 위해 10 mM 시트르산 완충액(pH 6.0)에서 10분간 처리 후 실온에서 20분간 냉각하는 것을 권장합니다. 항원 회복 후, 실온에서 10분 동안 1차 항체로 시료를 인큐베이션합니다. 양성 대조군으로는 피부 조직(skin tissue)을 권장합니다.
Target Information
Involucrin은 각질형성세포(keratinocyte)의 교차결합막(crosslinked envelope) 구성 요소로, 세포질에 존재하며 transglutaminase에 의해 막 단백질에 교차결합됩니다. 이 단백질은 각막을 포함한 다양한 중층편평상피(stratified squamous epithelia)에서 발현됩니다. 정상 표피에서는 상부 유극층에서 처음 발현되며, 배양된 각질형성세포에서는 기저층을 벗어난 모든 세포에서 발현됩니다. Involucrin 발현은 편평세포암종(squamous cell carcinomas) 및 전암성 병변에서 비정상적이며, 후두와 자궁경부의 중증 이형성증에서는 감소합니다. 해당 유전자는 1q21 위치에 있으며, calpactin I light chain, trichohyalin, profillaggrin, loricrin, calcyclin 등의 유전자와 함께 존재합니다.
