Product Specific Information
- Predicted to react with bovine based on sequence homology.
- Heat-mediated antigen retrieval recommended using 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
- After retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
- Suggested positive controls: tonsil, lymph node, or breast carcinoma.
Target Information
Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases. It contains a forkhead associated (FHA) domain that mediates association with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.
Expression pattern:
- Perinucleolar expression at G1
- Nuclear matrix expression at G2
- Chromosomal expression during M phase
Ki-67 serves as a proliferation marker since it is absent in G0 cells but increases from G1 through mitosis. It is useful for determining the growth fraction of neoplasms, with prognostic value comparable to the tritiated thymidine-labelling index. A low Ki-67 index correlates with low-grade tumors. Commonly used as a neuronal marker for cell cycling and proliferation.
