Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, involved in signal transduction and receptor regulation.
CD33 is expressed on myelomonocytic lineage cells, binds sialic acid residues on N- and O-glycans, and serves as a therapeutic target for acute myeloid leukemia (AML). It is present on granulocyte and macrophage precursors in bone marrow but absent on pluripotent stem cells. CD33 is also a useful marker for peripheral monocytes and for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
