Product Specific Information
- Overnight incubation is recommended.
- The epitope recognized by this antibody is reported to be sensitive to formaldehyde fixation and tissue processing.
- Acetone fixation for frozen sections is recommended.
- Suggested positive control for IHC: lymphatic tissue.
- For FACS analysis, use 10 µL of the suggested working dilution to label 1×10⁶ cells in 100 µL.
- Overnight incubation with primary antibody is recommended.
Target Information
CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2 and mediate receptor internalization.
CD33 is expressed on myelomonocytic lineage cells and binds sialic acid residues on cell surfaces. It serves as a therapeutic target for acute myeloid leukemia (AML) and is also expressed on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells.
As a marker for peripheral monocytes, CD33 helps distinguish myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
