Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IL-10. Anti-Human IL-10-specific antibody was purified by affinity chromatography employing an immobilized Human IL-10 matrix.

Sandwich ELISA:
To detect Human IL-10 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human IL-10 (500-P20Bt) as a detection antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Human IL-10.

Western Blot:
To detect Human IL-10 by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. Used in conjunction with compatible secondary reagents, the detection limit for Recombinant Human IL-10 is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes and lymphocytes. It plays a key role in immunoregulation and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. IL-10 enhances B cell survival, proliferation, and antibody production, blocks NF-kappa B activity, and regulates the JAK-STAT signaling pathway. Knockout studies indicate IL-10 is essential for intestinal immune regulation. Mutations are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis. Associated diseases include Graft-Versus-Host Disease and HIV-1.
