Product Specific Information

The eBio500A2 monoclonal antibody reacts with the 25 kD epsilon subunit of the mouse CD3 complex. CD3 subunits (gamma, delta, epsilon) are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells, as well as NKT cells.

Binding of eBio500A2 to CD3 initiates intracellular activation and proliferation. The 500A2 antibody partially cross-blocks 17A2 and 145-2C11 antibodies, indicating recognition of distinct but overlapping epitopes.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested with mouse splenocytes at ≤1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.

Super Bright 436 is excited with the violet laser line (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are structurally related immunoglobulin superfamily members encoded on chromosome 11.

CD3 components have long cytoplasmic tails with tyrosine-based motifs mediating signal transduction. CD3 is involved in TCR-induced growth arrest, survival, and proliferation.

CD3 antigen is expressed on:

• 68–82% of peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

Not expressed on B or NK cells. Reduced T lymphocytes may occur in autoimmune diseases. Defects in CD3 genes are associated with CD3 immunodeficiency.
