Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. Along with other CD3 subunits (gamma and delta), the epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. The SK7 and UCHT1 monoclonal antibodies cross-block binding, suggesting recognition of overlapping epitopes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage is 5 µL (0.5 µg) per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test.

Super Bright 436 can be excited with the violet laser line (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility before use.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into T-cell cytoplasm and regulating TCR surface expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits—members of the immunoglobulin superfamily encoded on chromosome 11. These subunits associate with signal transduction molecules via a double tyrosine-based motif. CD3 plays roles in TCR-induced growth arrest, survival, and proliferation.

CD3 antigen is present on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum. It is not expressed on B or NK cells. Decreased T lymphocyte percentages may occur in autoimmune diseases. Defects in the CD3 gene are associated with CD3 immunodeficiency.
