Product Specific Information
The OX35 monoclonal antibody reacts with rat CD4. CD4 is a co-receptor participating in T cell receptor signaling by interacting with MHC class II on antigen-presenting cells (APCs). It is expressed in thymic CD4+CD8+ double-positive and CD4+ single-positive thymocytes, as well as CD4+ helper T cells in the periphery. CD4 is a membrane glycoprotein with four extracellular immunoglobulin-like domains.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on rat splenocytes, ≤1.0 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Antibody should be titrated for optimal assay performance.
Super Bright 436 is excited by the violet laser (405 nm) and emits at 436 nm. Use a 450/50 bandpass filter or equivalent. Ensure instrument compatibility with this fluorochrome.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information
CD4 antigen is involved in MHC class II recognition and serves as a co-receptor for HIV. It is mainly expressed in T helper cells and other immune cells such as monocytes, macrophages, and dendritic cells. At the tissue level, CD4 is found in thymus, lymph nodes, tonsils, spleen, and certain regions of brain and gut. CD4 aids in T-cell activation via association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in infectious or immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
