Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
IL-1 alpha (Interleukin-1 alpha) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is encoded by the IL1A gene and signals through two receptors, IL-1RI and IL-1RII, both shared with IL-1 beta. The IL1A gene is located on chromosome 2q13.
IL-1 alpha plays a critical role in innate host defense by inducing other proinflammatory cytokines and initiating acute-phase responses. Its activity can be regulated by IL-1 Receptor Antagonist (IL-1RA), which blocks receptor binding through competitive inhibition.
This cytokine is involved in immune response, inflammation, and hematopoiesis. It is produced as a proprotein by monocytes and macrophages, processed upon cell injury, and induces apoptosis. IL-1 alpha regulates NF-κB and MAP kinase pathways, stimulates IL6 and COX-2 (PTGS2) expression, and enhances nitric oxide (NO) production. Elevated IL-1 alpha levels are associated with chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
