Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

IL-1 alpha (Interleukin-1 alpha) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is encoded by the IL1A gene and signals through two receptors, IL-1RI and IL-1RII, both shared with IL-1 beta.
Located on chromosome 2q13, IL-1 alpha plays a key role in innate host defense by inducing other proinflammatory cytokines and acute-phase responses.
Its activity is moderated by IL-1 Receptor Antagonist (IL-1RA), which blocks receptor binding through competitive inhibition.
IL-1 alpha regulates NF-kappaB and MAPK pathways, stimulates IL6 and COX-2 (PTGS2) expression, and enhances NO production.
High IL-1 alpha levels are linked to chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
