Product Specific Information
This TNKUPJ monoclonal antibody recognizes mouse CD163, a 130 kDa surface receptor expressed by subsets of tissue macrophages such as splenic red pulp macrophages, Kupffer cells, intestinal lamina propria macrophages, and a small fraction of peritoneal macrophages. Mouse monocytes do not express CD163, and unlike human CD163, it is not a strong M2 macrophage marker.
This antibody can detect intracellular CD163 in fixed and permeabilized cells. Since aggressive tissue dissociation may reduce surface CD163, intracellular staining is recommended in such cases.

Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily and serves as a receptor for the hemoglobin-haptoglobin complex. It protects tissues from oxidative damage and participates in iron recycling via endocytosis of hemoglobin/haptoglobin complexes.
CD163 is expressed on human monocytes and macrophages, upregulated by IL-10, IL-6, and dexamethasone, and shed from the cell surface upon LPS or PMA stimulation. It binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner and triggers intracellular signaling involving calcium mobilization and cytokine secretion (IL-6, CSF1).
