Product Specific Information

The monoclonal antibody OX41 reacts with rat CD172a, also known as signal-regulatory protein alpha (SIRPα).
SIRP family members are transmembrane glycoproteins with three Ig-like extracellular domains and a cytoplasmic tail containing two immunoreceptor tyrosine-based inhibition motifs (ITIMs).
CD172a is expressed by monocytes, macrophages, granulocytes, dendritic cells, and neurons.

SIRPα association with CD47 inhibits phagocytosis of CD47-presenting cells. Studies suggest that SIRPα ligation prevents phagocytosis by inactivating paxillin and cofilin, proteins involved in cytoskeleton remodeling.

Target Information

SIRPα (CD172a) is a receptor-type transmembrane glycoprotein expressed on myeloid-origin cells such as granulocytes, dendritic cells, macrophages, mast cells, and hematopoietic stem cells.
It acts as a substrate for activated tyrosine kinases (EGFR, PDGFR, src, insulin receptor) and negatively regulates receptor tyrosine kinase-coupled signaling pathways.

Ligand binding of SIRPα to CD47 results in phosphorylation of ITIMs within its cytoplasmic region, mediating recruitment and activation of tyrosine phosphatases SHP-1 and SHP-2.
This interaction inhibits macrophage-mediated phagocytosis and contributes to bi-directional activation of T cells and dendritic cells.
SIRPα regulates cellular responses to serum, growth factors, insulin, oncogenes, and adhesion, playing a role in physiological and pathological processes.
Cancer cells highly express CD47, activating SIRPα and inhibiting macrophage-mediated destruction of tumor cells.
