Product Specific Information

The eBioGHI/61 monoclonal antibody reacts with human CD163 (Haemoglobin Scavenger Receptor, M130), a member of the macrophage scavenger receptor family. CD163 is expressed by tissue macrophages, circulating blood monocytes, and possibly hematopoietic progenitor cells. It functions as a high affinity scavenger receptor for the hemoglobin-haptoglobin complex, facilitating clearance of hemoglobin from blood.

Upon ligand binding, CD163 triggers signal transduction leading to IL-6 and IL-10 production. Cytokines such as IL-6 and IL-10 can induce CD163 expression, while proinflammatory stimuli like LPS or TNF-α cause shedding of soluble CD163.

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein and receptor for the hemoglobin-haptoglobin complex. It protects tissues from hemoglobin-mediated oxidative damage and participates in iron recycling through endocytosis and heme breakdown.

CD163 is expressed on human monocytes and macrophages, especially in late inflammation phases. It is upregulated by IL-10, IL-6, and dexamethasone, and shed from the cell surface upon stimulation with LPS or PMA. Binding of hemoglobin/haptoglobin complexes is calcium- and pH-dependent, with higher affinity for multimeric haptoglobin complexes (HP1F phenotype).

CD163 signaling involves tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL6 and CSF1.
