The monoclonal antibody eBio5G3 recognizes CD171 (L1CAM), also known as neural cell adhesion molecule L1. CD171 is a member of the Ig superfamily containing six extracellular Ig domains and five fibronectin type III-like repeats. It functions as a cell adhesion molecule mediating homotypic and heterotypic cell–cell interactions in neuronal myelination, neurite outgrowth, and regeneration.

Expression of CD171 has been observed on monocytes, mature monocytic-derived and follicular dendritic cells, a minor subset of lymphocytes, neuronal tissue, and certain tumor cell lines. Expression on tumors is associated with tumor progression. The epitope of eBio5G3 is located in the amino-terminal Ig-like domain.

This antibody has been pre-titrated and validated by flow cytometric analysis using the tumor cell line Panc-1. Recommended use is 5 µL (0.06 µg) per test, defined as the amount of antibody sufficient to stain a cell sample in a final volume of 100 µL. Cell numbers may vary from 10⁵ to 10⁸ cells per test.

Target Information

L1CAM/CD171 is an axonal glycoprotein belonging to the immunoglobulin supergene family. Its ectodomain, composed of multiple immunoglobulin-like and fibronectin type III-like repeats, is linked via a single transmembrane sequence to a conserved cytoplasmic domain. This molecule plays a critical role in nervous system development, including neuronal migration and differentiation.

Mutations in the L1CAM gene cause several X-linked neurological syndromes collectively referred to as CRASH (corpus callosum hypoplasia, retardation, aphasia, spastic paraplegia, and hydrocephalus). Alternative splicing of a neuron-specific exon contributes to functional diversity.
