Product Specific Information

Description:
The 24-31 monoclonal antibody reacts with human CD154 (CD40 ligand, CD40L), a 39 kDa transmembrane glycoprotein in the TNF superfamily. It is transiently expressed by activated T cells and interacts with CD40 on antigen-presenting cells (B cells, monocytes/macrophages, dendritic cells), playing a key role in T-APC interactions and B-cell activation. The 24-31 clone cross-reacts with cynomolgus monkey CD154.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated for flow cytometry using stimulated normal human peripheral blood cells. Use 5 µL (0.125 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product # 00-822-49)
• 1-step Fix/Lyse Solution (Product # 00-5333-54)
  Store up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information

CD40 Ligand (CD40-L, CD154):
A membrane glycoprotein expressed on T cells that stimulates B cell proliferation and immunoglobulin secretion. It induces cytokine production and tumoricidal activity in monocytes and co-stimulates activated T cell proliferation, leading to IFN-γ, TNF-α, and IL-2 production.
