이 항체는 비인산화 Tau에 반응하는 항체를 제거하기 위해 비인산화 펩타이드로 음성 프리애드소브 처리되었으며, Ser262에서 인산화된 Tau 유래 펩타이드로 친화 크로마토그래피 정제를 통해 최종 생성되었습니다.
양성 대조군으로는 Recombinant human Tau ± PKA, 또는 human four repeat Tau와 SV40 small T antigen을 발현하는 CV-1 세포를 사용합니다.
Target Information
Tau는 신경세포 축삭에서 주로 발견되는 미세소관 결합 단백질로, tubulin 중합을 촉진하고 미세소관을 안정화시키는 역할을 합니다.
C-말단은 축삭 미세소관에 결합하고 N-말단은 신경세포막 구성 요소에 결합하여 Tau가 두 구조 사이의 연결 단백질로 작용함을 시사합니다.
Tau의 과인산화 형태는 Alzheimer’s disease(AD) 뇌에서 발견되는 paired helical filaments(PHF)의 주요 구성 요소입니다.
과인산화는 Tau의 미세소관 결합 기능을 손상시켜 미세소관 불안정화를 초래하고, 결국 신경세포 퇴행을 유발합니다.
Tau는 GSK-3β, PKA, cdk5, casein kinase II 등 다양한 세린/트레오닌 키나아제에 의해 인산화됩니다.
과인산화 Tau는 Alzheimer’s disease뿐 아니라 Pick’s disease, frontotemporal dementia, cortico-basal degeneration, progressive supranuclear palsy 등 다양한 신경퇴행성 질환에서 발견됩니다.
