Product Specific Information

PA1-774 detects rabphilin 3A from rat tissues.
It has been successfully used in Western blot procedures.
By Western blot, this antibody detects an ~75 kDa protein representing rabphilin 3A from rat brain whole protein extract.

The immunogen is a synthetic peptide corresponding to residues M(1)TDTVVNRWMYPGDGPLQ(18)C of rat Rabphilin 3A, a sequence completely conserved between rat and mouse rabphilin 3A.
The immunizing peptide (Cat. # PEP-102) is available for use in neutralization and control experiments.

Target Information

Rabphilin-3A contains two C2 domains (A and B) at the carboxy-terminal and a cysteine-rich region (residues 45–170) with two zinc finger motifs at the amino-terminal.
It belongs to a family of C-type tandem C2 proteins, including synaptotagmins and Doc2.
Rabphilin is expressed in neuroendocrine cells and co-localizes with Rab3A on synaptic vesicles and chromaffin granules.
It binds Rab3a/GTP/Mg²⁺ within amino-terminal residues 45–170, and binds calcium ions and phosphatidylinositol 4,5-bisphosphate-containing lipid vesicles within its C2 domains.
Rabphilin-3A acts as a positive regulator of calcium-dependent exocytosis, while Rab3a acts as a negative regulator.
Although they associate, their effects on exocytosis appear to be independent, likely mediated by an unknown factor recognizing the Rab3 binding domain.
