Product Specific Information

The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and is essential for T cell development and function. Binding of RM4-5 is blocked by GK1.5.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Tested on mouse splenocytes; use ≤0.125 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimize concentration for best performance.
  • Fluorochrome: Super Bright 600 (excitation 405 nm, emission 600 nm; violet laser compatible). Use 610/20 bandpass filter.
  • Buffer Recommendation: When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions.
  • Light Sensitivity: Protect from light to prevent photo-induced oxidation.
  • Fixation: Samples can be stored up to 3 days at 2–8°C in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness.
  • Laser: Violet Laser (405 nm excitation, 600 nm emission)

Target Information

CD4 antigen is a co-receptor for MHC class II molecules and is involved in T-cell activation. It is primarily expressed in T helper cells, and also in monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 functions to initiate or enhance T-cell activation through association with the T-cell receptor complex and protein tyrosine kinase Lck. It may also play a role in neuronal damage in infectious and immune-mediated CNS diseases.
