Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa surface receptor expressed by thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and is essential for T cell development and function. GK1.5 binding is blocked by RM4-5.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometry of mouse splenocytes at ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited at 405 nm (violet laser) and emits at 600 nm. Recommended detection filter: 610/20 bandpass. Protect from light due to photo-induced oxidation sensitivity.

Staining Recommendations:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

CD4 is a co-receptor for MHC class II recognition and plays a key role in T-cell activation via association with the T-cell receptor complex and Lck kinase. It is expressed on T helper cells, monocytes, macrophages, and dendritic cells, and is found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. CD4 may also mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
