Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a key role in cellular response to genotoxic stress and is a major transcriptional target of the p53 protein. In the nucleus, p21 binds and inhibits CDK1 and CDK2, blocking cell cycle progression from G1 to S phase or G2 to mitosis after DNA damage, allowing DNA repair. In the cytoplasm, p21 exhibits anti-apoptotic effects by inhibiting caspase 3 and apoptotic kinases such as ASK1 and JNK. Thus, p21 has a dual role in carcinogenesis, functioning both as a tumor suppressor and as an oncogene depending on cellular context.
