Product Specific Information
The 6B10.2 monoclonal antibody reacts with human CD247 (CD3 zeta chain). The CD3 complex consists of gamma, delta, epsilon, and zeta chains, forming part of the TCR/CD3 complex. CD3 zeta is expressed on immature and mature thymocytes, T lymphocytes, and NK cells. It plays a key role in T cell activation via tyrosine phosphorylation of ITAM motifs.
This antibody recognizes a transmembrane epitope of the CD3 zeta chain; therefore, intracellular staining is required for flow cytometric analysis. It detects the zeta chain as a homodimer but not as a monomer.
Applications Reported:
Intracellular staining followed by flow cytometric analysis.
Applications Tested:
Pre-titrated and tested using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) following BestProtocols® Protocol A.
Use 5 µL (0.03 µg) per test in 100 µL volume, with 10⁵–10⁸ cells/test.
Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm, excited by the 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable). Protect from light due to photo-induced oxidation sensitivity.
Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.

Target Information
The CD3z complex is essential for transducing antigen recognition signals into T cells and regulating TCR complex expression. CD3 subunits (gamma, delta, epsilon, zeta) are encoded on chromosome 11 and contain ITAM motifs for signal transduction. CD3 is expressed on most T lymphocytes and thymocytes, but not on B or NK cells. Altered CD3 expression is associated with immune deficiencies such as Immunodeficiency 25 and T-B+ Severe Combined Immunodeficiency due to CD3δ/ε/ζ defects.
