The monoclonal antibody C9.1 reacts with mouse CD244.1, a receptor expressed on NK cells, γδ T cells, some CD8+ T cells, basophils, and monocytes in most inbred mouse strains except C57BL/6 and C58, which express the CD244.2 allotype.

CD244 is a CD2-related receptor belonging to the SLAM family and exists as two isoforms with opposing functions. It associates with CD48, regulating NK cell proliferation, cytotoxicity, and cytokine production. The CD244/CD28 interaction inhibits NK cytotoxicity, contributing to self-tolerance. CD244 may enhance or inhibit CD8+ T cell effector function depending on SAP levels.

The C9.1 antibody has been reported to have antagonist activity on activated NK cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of BALB/c mouse splenocytes at ≤0.125 µg/test (100 µL final volume). Cell number should be determined empirically (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm). It can replace PerCP-Cyanine5.5. Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

This gene encodes a cell surface receptor expressed on NK cells and some T cells, mediating non-MHC restricted killing. Interaction between NK and target cells via this receptor modulates NK cytolytic activity. Alternatively spliced variants encoding different isoforms have been identified.
