This 7-3 (7) monoclonal antibody reacts with human CD258 (LIGHT), a 29-kDa type II transmembrane protein of the TNF ligand superfamily related to lymphotoxin-β (LT-β).
LIGHT is expressed on activated T cells, particularly CD8+ T cells, and plays roles in cell survival, inflammation, and tumor eradication. Upon binding to herpesvirus entry mediator (HVEM), LIGHT induces a costimulatory effect enhancing T cell proliferation and cytokine production. It can also interact indirectly with the LT-β receptor.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested on stimulated normal human peripheral blood cells at 5 µL (0.125 µg) per test.
A test is defined as the amount of antibody that stains a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test.

PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm). It can substitute for PerCP-Cyanine5.5. Recommended filter: 710/50 bandpass (695/40 acceptable).

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

Apoptosis (programmed cell death) occurs during normal differentiation and development. It is induced by cytokines such as TNF and Fas ligand via death domain-containing receptors (TNFR1, Fas).
The death domain (DD)-containing adapter molecule Daxx (death associated protein 6) binds specifically to the Fas death domain, enhances Fas-induced apoptosis, and activates the JNK pathway.
Daxx is widely expressed in fetal and adult tissues, indicating its important role in Fas signaling pathways.
