Product Specific Information

Description:
The 93 monoclonal antibody reacts with an epitope shared by mouse CD16 and CD32. These are low affinity receptors (FcγRIII and FcγRII) for the IgG Fc portion and are expressed on B cells, monocytes/macrophages, NK cells, and neutrophils.

Applications Reported:
The 93 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.25 µg per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best results.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-54)
  Clone-specific performance should be determined empirically.

Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD16 (FcγRIII) is a 50–65 kDa cell surface molecule expressed in two forms:

• Transmembranous form on NK and some T cells
• Phosphatidylinositol-linked form on granulocytes

It mediates antibody-dependent cell-mediated cytotoxicity (ADCC).

CD32 (FcγRII) is a 40 kDa glycoprotein expressed on B cells, monocytes, granulocytes, and platelets.
It mediates endocytosis, secretion, cytotoxicity, and immunomodulation, and is involved in immune complex phagocytosis and antibody regulation.
Variants of CD32 may increase susceptibility to systemic lupus erythematosus (SLE).
