Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein. CD14 is expressed on monocytes, interfollicular macrophages, and some dendritic cells. LPS-LBP complexes bind with high affinity to monocytes via surface CD14.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (1.0 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
• Light Sensitivity: Protect from light; tandem dye is sensitive to photo-induced oxidation.
• Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer or 1-step Fix/Lyse Solution with minimal brightness loss.
• Excitation / Emission: 488–561 nm / 775 nm; compatible lasers: Blue, Green, Yellow-Green.
• Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on mature monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide receptor and is released into serum as soluble sCD14. CD14 interacts with LBP to bind LPS, discriminates structural LPS differences, and plays roles in immune and inflammatory responses. Associated diseases include Mycobacterium chelonae infection and croup.
