Product Specific Information
The eBioCB16 monoclonal antibody recognizes CD16 (FcγRIII), a low-affinity receptor for IgG with an apparent molecular weight of 50–80 kDa.
CD16 includes two genes:
- CD16A (FcγRIIIA): Expressed on macrophages and NK cells (transmembrane form)
- CD16B (FcγRIIIB): Expressed on neutrophils (GPI-anchored form)
Polymorphisms (NA-1, NA-2) in CD16B affect phagocytic capacity. CD16 binds IgG immune complexes, showing preference for IgG1 and IgG3. Upon binding, it triggers signaling cascades leading to ADCC, phagocytosis, degranulation, and proliferation.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry using normal human peripheral blood cells.
Use at 5 µL (0.03 µg) per test in a final volume of 100 µL.
Cell number: 10⁵–10⁸ cells/test (empirically determined).
Light Sensitivity: Protect from light (photo-oxidation sensitive).
Fixation: Samples can be stored in:
- IC Fixation Buffer (Product #00-822-49)
- 1-step Fix/Lyse Solution (Product #00-5333-54)
for up to 3 days at 4°C in the dark with minimal signal loss.
Excitation: 488–561 nm
Emission: 775 nm
Compatible Lasers: Blue, Green, Yellow-Green
Filtration: 0.2 µm post-manufacturing filtered
Target Information
CD16 (FcγRIII) encodes a receptor recognizing the Fc portion of IgG, involved in immune complex clearance and antibody-dependent cellular cytotoxicity (ADCC).
- Gene: FCGR3A (chromosome 1)
- Expression: NK cells (transmembrane glycoprotein), neutrophils (PI-linked form via FCGR3B)
- Clinical relevance: Mutations linked to Immunodeficiency 20, recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia.
- Variants: Multiple alternatively spliced isoforms exist.
