Product Specific Information

This 1RNM44N monoclonal antibody recognizes mouse CD170 (Siglec F), a cell surface lectin in the Ig superfamily that binds sialic acid-containing glycoconjugates. The cytoplasmic domain contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) initiating intracellular signaling upon ligand binding.
Siglec F is primarily expressed on eosinophils and alveolar macrophages, with lower levels on immature myelomonocytic cells. It is the functional ortholog of human Siglec 8 but is not expressed on mast cells in mice.
This antibody recognizes a formaldehyde-fixed epitope.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse thioglycolate-elicited peritoneal exudate cells at ≤0.125 µg/test (100 µL final volume). Cell numbers can range from 10⁵–10⁸ cells/test. Titrate for optimal performance.

Fluorochrome Information:
Alexa Fluor™ 700 emits at 723 nm, excited by the red laser (633 nm). Recommended optical setup: 685 LP mirror and 710/20 filter.
Excitation: 633–647 nm; Emission: 723 nm; Laser: Red Laser.
Post-manufacturing filtration: 0.2 µm.


Target Information

SIGLEC5 is a putative adhesion molecule mediating sialic acid-dependent binding. It interacts with both α2,3- and α2,6-linked sialic acids, possibly masked by cis interactions on the same cell surface. Expressed in monocytic/myeloid lineage cells, SIGLEC5 is found at high levels in peripheral blood leukocytes, spleen, and bone marrow, with lower expression in lymph node, lung, appendix, placenta, pancreas, and thymus. It is expressed in monocytes and neutrophils but absent in early leukemic myelomonocytic cell lines.
