Description:
The eBioC1.7 monoclonal antibody reacts with human CD244 (2B4, p38), a 38 kDa protein expressed on NK cells, subsets of CD8⁺ T cells, γδ T cells, monocytes, basophils, and eosinophils. CD244 binds to CD48, resulting in NK cell activation and enhanced cytotoxicity. C1.7 binding induces tyrosine phosphorylation and SAP recruitment. In X-linked lymphoproliferative disorder (XLPD), SAP mutations impair this interaction.

Applications Reported:

• Reported for use in flow cytometric analysis.

Applications Tested:

• Pre-diluted and tested by flow cytometry on normal human peripheral blood cells.
• Use 5 µL (0.4 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, add 5 µL per 100 µL of cell sample.

Target Information

CD244 (2B4, SLAMF4) is a cell surface receptor on NK cells and some T cells that mediates non-MHC-restricted cytotoxicity. Interaction with SLAMF2 activates both SLAMF4- and SLAMF2-expressing cells. Reduced SLAMF4 levels are observed in systemic lupus erythematosus, implicating CD244 in autoimmune disease pathology. CD244 dysfunction is associated with rheumatoid arthritis and lymphoproliferative syndromes. Multiple transcript variants exist.
