Product Specific Information

Description:
The eBioC1.7 monoclonal antibody reacts with human CD244 (2B4, p38). CD244 is a 38 kDa protein expressed on NK cells, subsets of CD8+ T cells, γδ T cells, monocytes, basophils, and eosinophils. Binding of its ligand CD48 results in NK cell activation and enhanced cytotoxicity. The C1.7 antibody binds the same residue in the V domain as CD48, inducing NK cell activation via tyrosine phosphorylation and recruitment of SAP (SLAM-associated protein). Mutations in SAP are associated with X-linked lymphoproliferative disorder (XLPD).

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Use 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Staining Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample or per 100 µL of cell sample (10³–10⁸ cells) for single-color controls.

Target Information

CD244 (2B4, SLAMF4):
A cell surface receptor expressed on NK cells and some T cells, mediating non-MHC restricted cytotoxicity. CD244 interacts with SLAMF2, activating both SLAMF4- and SLAMF2-expressing cells. Reduced SLAMF4 expression has been observed in systemic lupus erythematosus, suggesting a role in autoimmune pathology. CD244 dysfunction is associated with rheumatoid arthritis and lymphoproliferative syndromes. Multiple alternatively spliced transcript variants exist.
