Product Specific Information

The eBioC1.7 monoclonal antibody reacts with human CD244 (2B4, p38), a 38 kDa protein expressed on NK cells, CD8+ T cells, γδ T cells, monocytes, basophils, and eosinophils.
Binding of CD48 to CD244 activates NK cells, enhancing cytotoxicity and IFN-γ secretion. The C1.7 clone binds the same residue as CD48, inducing NK cell activation through tyrosine phosphorylation and SAP recruitment.

Patients with X-linked lymphoproliferative disorder (XLPD) have SAP mutations preventing CD244 binding.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested on human peripheral blood cells. Recommended 5 µL (0.4 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample when using antibody cocktails.
• For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology for engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD244 (2B4, SLAMF4) is a receptor on NK and some T cells mediating non-MHC restricted killing.
Interaction with SLAMF2 modulates NK-cell activity.
Lower SLAMF4 expression in lupus patients suggests involvement in autoimmune pathology.
Associated diseases: rheumatoid arthritis, lymphoproliferative syndrome.
Alternative splicing generates multiple isoforms.
