Product Specific Information

The eBioC1.7 monoclonal antibody reacts with human CD244 (2B4, p38), a 38 kDa protein expressed on NK cells, CD8+ T cells subset, γδ T cells, monocytes, basophils, and eosinophils. CD244 ligand CD48 binding activates NK cells, enhancing cytotoxicity and IFN-γ secretion. C1.7 antibody binding induces tyrosine phosphorylation and recruitment of SAP (SLAM-associated protein). In X-linked lymphoproliferative disorder (XLPD) patients, SAP mutation prevents binding to phosphorylated CD244.

• Recommended use: 5 µL (0.6 µg) per test in 100 µL final volume; cell count 10⁵–10⁸ cells/test

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample before mixing with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD244 (2B4, SLAMF4) is a receptor on NK cells and some T cells that mediates non-MHC restricted cytotoxicity. Interaction with SLAMF2 modulates NK-cell activity. Reduced SLAMF4 expression is observed in systemic lupus erythematosus, suggesting a role in autoimmune pathology. CD244 dysfunction is associated with rheumatoid arthritis and lymphoproliferative syndrome. Multiple transcript variants encoding different isoforms have been identified.
