Product Specific Information

This eyedio monoclonal antibody reacts with human indoleamine-2,3-dioxygenase (IDO, INDO, IDO1), an intracellular enzyme that catalyzes the degradation of tryptophan to kynurenines.
IDO is expressed in multiple tissues and cell types, including macrophages, plasmacytoid dendritic cells, and various cell lines. It can be induced by IFN-gamma or other inflammatory stimuli.
IDO expression in antigen-presenting cells and tumors contributes to immune suppression by tryptophan depletion and production of metabolites that induce T cell apoptosis and regulatory T cell formation.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and validated using Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00). Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and is compatible with cytometers equipped with a 355 nm UV laser. Ensure instrument capability before use.

Buffer Recommendations:
For panels containing multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Target Information

IDO1 is an intracellular heme-containing enzyme that catalyzes the oxidative cleavage of the indole ring of molecules such as tryptophan, serotonin, and melatonin, producing biologically active kynurenines.
IDO1 is expressed in human tissues, macrophages, and dendritic cells. IFN-induced IDO1 activation contributes to antimicrobial defense and immunoregulation.
Tryptophan degradation by IDO1 inhibits viral, bacterial, and parasitic growth and plays roles in infection, pregnancy, autoimmunity, transplantation, and cancer.
