Product Specific Information

The XMG1.2 antibody reacts with mouse interferon (IFN) gamma and functions as a neutralizing antibody. IFN gamma is a 20 kDa cytokine produced by activated T, B, and NK cells with antiviral and antiparasitic activity. It acts synergistically with cytokines such as TNF alpha to inhibit proliferation of normal and transformed cells. Glycosylation does not affect its biological activity.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested for intracellular staining of mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended ≤0.5 µg/test. Optimal titration is advised.

Fluorochrome Information:
Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and is designed for use with cytometers equipped with an ultraviolet (355 nm) laser.

When using multiple polymer dye-conjugated antibodies (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Target Information

IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor produced primarily by T-lymphocytes and NK cells in response to antigens and cytokines. It is a dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits. IFN gamma exhibits species-specific biological activity and does not cross-react between human and mouse.

Functions include:

  • Antiviral and tumor antiproliferative activity
  • Induction of MHC class I and II expression
  • Macrophage activation
  • Enhanced immunoglobulin secretion by B cells

Activation occurs via IFN gamma receptor I and II through the JAK-STAT pathway.
Expression is upregulated by IL2, FGF basic, EGF, and downregulated by vitamin D3 or DMN.
Mutations in the IFN gamma gene are associated with aplastic anemia.
