Product Specific Information

The SHLR17 monoclonal antibody reacts with human interleukin (IL)-17F, a 34 kDa homodimeric member of the IL-17 family. IL-17F and IL-17A are mainly produced by Th17 cells and have overlapping but distinct functions. IL-17F binds to IL-17RA/IL-17RC receptor complexes, inducing secretion of pro-inflammatory cytokines and chemokines, and recruiting neutrophils.

Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and tested using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended use at 5 µL (0.5 µg) per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and requires a cytometer equipped with a 355 nm UV laser.
Excitation: 350 nm; Emission: 395 nm; Laser: Ultraviolet.

Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Target Information

IL-17F (Interleukin 17F, CTLA-8) is a cytokine produced by activated T cells, mast cells, and basophils. It plays a role in allergic airway inflammation and induces cytokines, chemokines, and adhesion molecules in various cell types. IL-17F acts via IL-17R to promote monocyte and neutrophil recruitment and stimulate pro-inflammatory cytokines such as TNFα, IL-1β, IL-6, and IL-8. It is implicated in chronic inflammatory diseases like rheumatoid arthritis, psoriasis, and multiple sclerosis. IL-17F also inhibits endothelial angiogenesis and induces IL2, TGFB1, and MCP-1 production.
