Product Specific Information

The eBioA1 monoclonal antibody reacts with human CD39 (ENTPD1 or NTPDase), an integral membrane ectoenzyme with two transmembrane domains forming a homotetramer. CD39 removes toxic extracellular ATP by converting it to ADP or AMP, functioning together with CD73. It is expressed on activated lymphocytes, subsets of T cells, B cells, dendritic cells, and weakly on monocytes and granulocytes.

CD39 and CD73 are expressed on regulatory T cells (Treg), aiding their migration into inflamed areas with high ATP levels. Expression on Foxp3+CD4+ cells ranges from 25–45%.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis and immunohistochemical staining.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Recommended viability dyes: Invitrogen LIVE/DEAD Fixable Dead Cell stains.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample prior to combining with cells.
  • For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD39 is a 70–100 kD cell surface antigen expressed on peripheral blood B cells, monocytes, T cell clones, and weakly on granulocytes. It has intrinsic ecto-ATPase activity and is upregulated on activated T and B cells.
