Product Specific Information

The eBioA1 monoclonal antibody reacts with human CD39, also known as ectonucleoside triphosphate diphosphohydrolase 1 (ENTPD1) or NTPDase. CD39 is an integral membrane protein with two transmembrane domains and exists as a homotetramer. It is the most prominent ectoenzyme of the immune system, functioning to remove toxic extracellular ATP by converting it to ADP or AMP. CD39 works together with CD73 to hydrolyze ATP and is well characterized on Langerhans cells.

Expression of CD39 was originally identified on activated lymphocytes and is also found on subsets of T cells, B cells, and dendritic cells, as well as weak staining on monocytes and granulocytes. CD39 and CD73 are expressed on regulatory T cells (Treg), facilitating their entry into inflamed areas with high ATP levels. Expression on Foxp3+CD4+ cells ranges from 25–45%.

Applications Reported

This eBioA1 (A1) antibody is suitable for flow cytometric analysis.

Applications Tested

Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.6 µg) per test. A test is defined as staining a cell sample in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.

Compatibility Notes

NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal results.

CellBlox Plus Blocking Buffer

This NovaFluor conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which contains formulation improvements over the previous version.

  • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
  • For single-color controls: use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor Technology

NovaFluor conjugates utilize Phiton™ technology based on novel nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover and spread.

Target Information

CD39 is a 70–100 kDa cell surface molecule expressed on peripheral blood B cells, monocytes, T cell clones, and weakly on granulocytes. It has intrinsic ecto-ATPase activity and is induced on T cells and increased on B cells as a late activation antigen.
