Product Specific Information

Description:
The eBioA1 monoclonal antibody reacts with human CD39, also known as ectonucleoside triphosphate diphosphohydrolase 1 (ENTPD1) or NTPDase. CD39 is an integral membrane protein with two transmembrane domains and exists as a homotetramer. It is the most prominent ectoenzyme of the immune system, functioning to remove extracellular ATP by converting it to ADP or AMP. CD39 works with CD73 to hydrolyze ATP and is well characterized on Langerhans cells. Expression is observed on activated lymphocytes, subsets of T and B cells, dendritic cells, and weakly on monocytes and granulocytes. CD39 and CD73 are also found on regulatory T cells (Treg), aiding their migration into inflamed areas with high ATP levels. Expression of CD39 on Foxp3⁺CD4⁺ cells ranges from 25–45%.

Applications Tested:

  • Pre-titrated and validated for flow cytometry using normal human peripheral blood cells
  • Recommended usage: 5 µL (0.4 µg) per test (100 µL final volume)
  • Cell number: 10⁵–10⁸ cells/test
  • Optimize titration for best performance

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
  • Always use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) with NovaFluor conjugates for optimal staining.
    • Add 5 µL per sample (regardless of number of NovaFluors).
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures that enable engineered fluorescent signatures with reduced spillover and spread.

Target Information

CD39 is a 70–100 kDa cell surface antigen expressed on peripheral blood B cells, monocytes, T cell clones, and weakly on granulocytes. It has intrinsic ecto-ATPase activity, and its expression is induced on T cells and increased on B cells as a late activation antigen.
