- P300A was successfully used to detect human TNF alpha in ELISA application.
- This antibody is predicted to react with mouse, rat, and non-human primate based on sequence homology.
- Intact IgG appears on a non-reducing gel as ~150 kDa band and upon reduction generates a ~25 kDa light chain band and a ~50 kDa heavy chain.
- Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning the specific antibody DNA sequences from immunoreactive rabbits.
- Advantages include better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity due to the larger rabbit immune repertoire.
Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It exists as multimers but shows a single 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies suggest a neuroprotective role and tumor necrosis activity. TNF alpha participates in immune responses to bacterial, viral, parasitic, and fungal infections, and contributes to tumor cytolysis. It is implicated in autoimmune diseases, insulin resistance, and cancer.
