Product Specific Information

The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells and plays a critical role in T cell development and function. In T cells, CD4 associates with the tyrosine kinase p56lck through its cytoplasmic tail. Binding of RM4-5 is blocked by GK1.5.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Tested on mouse thymocytes and splenocytes at ≤ 0.2 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.
• Compatibility: NovaFluor dyes are not compatible with DNA-intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Buffer Recommendation: Ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), required for optimal staining. Add 5 µL per sample (for any number of NovaFluors). For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).
• Technology: NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD4 antigen participates in recognition of MHC class II molecules and serves as a co-receptor for HIV. It is expressed mainly on T helper cells, and also on monocytes, macrophages, and dendritic cells. At the tissue level, CD4 is found in thymus, lymph nodes, tonsils, spleen, and certain regions of the brain and gut. CD4 aids in early T-cell activation via association with the T-cell receptor complex and kinase Lck, and may play a role in neuronal damage in infectious or immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
