Product Specific Information
The XMG1.2 antibody reacts with mouse interferon (IFN) gamma and is a neutralizing antibody. Mouse IFN gamma is a 20 kDa factor produced by activated T, B, and NK cells, exhibiting antiviral and antiparasitic activity. It synergizes with cytokines such as TNF alpha to inhibit cell proliferation. Glycosylation does not affect its biological activity.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes using Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended ≤ 0.5 µg per test (100 µL final volume). Cell number: 10^5–10^8 cells/test. Antibody titration recommended for optimal performance.
Fluorochrome Information: Brilliant Ultra Violet™ 661 (BUV661) emits at 660 nm and is designed for UV laser (355 nm) cytometers.
Use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific polymer interactions.
Light-sensitive: protect vial and samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information
IFN gamma (Interferon gamma, Type II interferon) is produced mainly by T-lymphocytes and NK cells. It is a dimeric glycoprotein (~45 kDa) with antiviral and antiproliferative activity, inducing MHC class I and II expression, macrophage activation, and immunoglobulin secretion. IFN gamma acts via IFN gamma receptor I/II and the JAK-STAT pathway. Human IFN gamma does not cross-react with mouse IFN gamma. Expression is upregulated by IL2, FGF basic, EGF, and downregulated by vitamin D3 or DMN. Mutations are associated with aplastic anemia.
