Product Specific Information

Description:
The 4S.B3 monoclonal antibody reacts with interferon-gamma (IFNγ), a 17 kDa factor produced by activated T and NK cells with antiviral and antiparasitic activity. IFNγ synergizes with TNFα to inhibit cell proliferation and modulate immune responses through high-affinity receptors.

Applications Reported:
Suitable for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-diluted and validated for intracellular staining of human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00). Recommended use: 5 µL (0.5 µg) per test (100 µL final volume).

Fluorochrome Information:
Brilliant Ultra Violet™ 661 (BUV661) emits at 660 nm and is designed for cytometers with a 355 nm UV laser.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to avoid photo-induced oxidation.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54).

Target Information

IFN gamma (Interferon gamma, Type II interferon):
A macrophage activation factor produced by T-lymphocytes and NK cells in response to antigens and cytokines. It functions as a dimeric glycoprotein (~45 kDa) with antiviral, antiproliferative, and immunomodulatory activities. IFNγ induces MHC I/II expression, activates macrophages, and enhances immunoglobulin secretion.

Human IFNγ shows species specificity, with no cross-reactivity to mouse IFNγ. It is regulated by IL-2, FGF basic, and EGF, and downregulated by vitamin D3 or DMN. Mutations in the IFNγ gene are associated with aplastic anemia.
