Product Specific Information

The 22URTI monoclonal antibody reacts with human interleukin (IL)-22, a 20 kDa cytokine of the IL-10 family secreted mainly by Th17 and NK cells. IL-22 expression is induced by IL-23 and acts through a heterodimeric receptor composed of IL-10R2 and IL-22R1.

Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and validated using human peripheral blood cells with the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 661 (BUV661) is a tandem dye emitting at 660 nm, excited by a 355 nm UV laser. Ensure the cytometer is equipped for UV detection.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light; dye is sensitive to photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54).

Target Information

IL-22, also known as IL-10-related T-cell derived inducible factor, is an alpha-helical cytokine of the IFN-IL-10 family. It is produced mainly by activated T cells and NK cells and acts through the IL-22R1/IL-10R2 receptor complex, activating the JAK-STAT pathway. IL-22 influences nonimmune cells such as keratinocytes, hepatocytes, and epithelial cells, regulating innate immunity, wound healing, and inflammatory responses. It is co-expressed with IL-17A and IL-17F in Th17 cells and plays a role in host defense and tissue repair.
