Description:
The TL2.1 monoclonal antibody reacts with human Toll-like receptor 2 (TLR2). TLR family members are type I transmembrane proteins with leucine-rich extracellular domains and cytoplasmic domains homologous to the type I IL-1 receptor. TLR2 and TLR4 act as pattern recognition receptors involved in innate immunity and inflammation. TLR2 is expressed by peripheral blood monocytes and recognizes diverse pathogens. TL2.1, a blocking antibody, is used to study TLR2-mediated cytokine production in human PBMCs. It can immunoprecipitate human TLR2 (~90 kDa) from PBMC and HMEC.

Applications Reported:
Used for flow cytometry, immunoprecipitation, immunoblotting (WB), and immunohistochemistry (frozen and paraffin sections). Also reported for blocking TLR2-mediated cytokine production. Functional Grade TL2.1 (Product #16-9922-82) is recommended for functional assays.

Applications Tested:
Tested by flow cytometry using normal human peripheral blood cells. Recommended ≤ 2 µg/test (100 µL final volume). Optimize antibody concentration per assay.

Target Information

TLR2 is a Toll-like receptor that plays a key role in pathogen recognition and activation of innate immunity. It signals via adaptor molecules such as MyD88 and TIRAP to activate kinases and transcription factors including PKCα/β and NF-κB. TLR2 forms heterodimers with TLR1 or TLR6 to recognize bacterial and mycoplasma lipoproteins. It is highly expressed in peripheral blood leukocytes and mediates responses to Gram-positive bacteria and yeast. TLR2 expression is regulated by immune modulators such as LPS, GM-CSF, IL-1, IL-10 (upregulation), and IL-4, IFN-γ, TNF (downregulation).
