Product Specific Information
The HTA125 monoclonal antibody reacts with human Toll-like receptor 4 (TLR4). TLR4, a member of the Toll family, is a type I transmembrane protein involved in innate immunity and inflammation. It forms a complex with MD-2 and CD14 for LPS recognition and signaling. TLR4 is expressed on peripheral blood monocytes and shows variable expression among donors.
For optimal staining of TLR4, use the lysed whole blood protocol rather than Ficoll-gradient prepared cells, as the latter reduces staining intensity.
Applications Reported: Flow cytometric analysis, immunoprecipitation, and blocking of LPS-induced cytokine production.
Applications Tested: Flow cytometric analysis of normal human peripheral blood cells (≤1 µg/test).
Purity: >90% (SDS-PAGE)
Aggregation: <10% (HPLC)

Target Information
TLR4 is a Toll-like receptor involved in pathogen recognition and activation of innate immunity. It is expressed mainly in placenta and myelomonocytic leukocytes. TLR4 participates in LPS-induced signal transduction with CD14 and LY96. Mutations in TLR4 affect LPS responsiveness. It is expressed by peripheral blood monocytes, a subset of B-cells, and placenta. TLR4-deficient mice show increased susceptibility to Gram-negative sepsis.
