Product Specific Information

Description:
The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by thymocytes, T-helper cells, and monocytes. CD4 serves as a receptor for HIV. The OKT4 antibody recognizes a distinct epitope from RPA-T4 and does not cross-block its binding.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.125 µg) per test in a final volume of 100 µL. Cell number can range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Alexa Fluor™ 700 emits at 723 nm and can be excited with a red laser (633 nm). Recommended optical setup: 685 LP mirror and 710/20 filter.

Target Information

CD4 antigen is involved in MHC class II recognition and functions as a co-receptor for HIV. Primarily expressed in T-helper cells, CD4 is also found in monocytes, macrophages, and dendritic cells. Tissue-level expression includes thymus, lymph nodes, tonsils, spleen, and certain regions of the brain and gut. CD4 assists in T-cell activation through association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in infectious or immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
