Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, a subpopulation of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays a crucial role in T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse thymocyte and splenocyte suspensions. Recommended ≤ 0.125 µg per test.
A test is defined as the amount of antibody that stains a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Fluorochrome Information:
Alexa Fluor® 700 emits at 723 nm and is excited with the red laser (633 nm). Most instruments require a 685 LP mirror and 710/20 filter.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. CD4 is mainly expressed on T helper cells but also found on monocytes, macrophages, and dendritic cells. Tissue-level expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 functions to initiate or augment early T-cell activation via association with the T-cell receptor complex and protein tyrosine kinase Lck. It may also mediate neuronal damage in infectious and immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
