Product Specific Information

  • For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

The protein encoded by the classic MBP gene is a major constituent of the myelin sheath of oligodendrocytes and Schwann cells in the nervous system. MBP-related transcripts are also present in bone marrow and the immune system. These arise from the long MBP gene (also known as Golli-MBP) that contains three additional exons upstream of the classic MBP exons.
Alternative splicing from the Golli and MBP transcription start sites gives rise to two sets of MBP-related transcripts and gene products:

  1. Golli-MBP transcripts — contain three unique exons spliced in-frame to one or more MBP exons, encoding hybrid proteins with N-terminal Golli and MBP sequences.
  2. Classic MBP transcripts — contain only MBP exons and produce the well-characterized myelin basic proteins.

This complex gene structure is conserved among species, suggesting that the MBP transcription unit is an integral part of the Golli transcription unit and that this arrangement is important for gene regulation and function.
