Product Specific Information

Description:
The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4+ T cells. TNF alpha has cytolytic activity against tumor cells and plays a critical role in immune regulation. It can form dimers and trimers and also exists as a 26 kDa membrane-bound form. The MAb11 antibody is a neutralizing antibody.

Applications Reported:
Reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and corresponding protocol. Recommended usage is 5 µL (0.25 µg) per test (100 µL final volume). Cell number per test may range from 10⁵ to 10⁸ depending on experimental conditions.

Fluorochrome Information:
Alexa Fluor® 700 emits at 723 nm and is excited with the red laser (633 nm). Recommended instrument setup includes a 685 LP mirror and 710/20 filter.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune response, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It exists as multimers but shows a 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies in mice indicate neuroprotective roles and tumor necrosis activity. TNF alpha contributes to immune responses against infections and tumor cytotoxicity, often synergistic with interferon-gamma. It is implicated in autoimmune diseases, insulin resistance, and cancer.
