Product Specific Information

This antibody recognizes CD40, a type I transmembrane glycoprotein belonging to the tumor necrosis factor receptor (TNFR) superfamily.
CD40 is broadly expressed on antigen-presenting cells (APCs) such as dendritic cells, B cells, macrophages, and monocytes, as well as non-immune endothelial and epithelial cells, and various tumors.
Upon binding to its ligand CD154, CD40 acts as a costimulatory molecule for the activation of B cells, dendritic cells, monocytes, and other APCs.


Target Information

The CD40 antigen is a single chain glycoprotein and a member of the TNF/nerve growth factor superfamily, showing significant homology to the Hodgkin’s disease-associated antigen CD30.
CD40 is expressed on all B cells except plasma cells and is found on epithelial cells, carcinomas, and lymphoid dendritic cells.
It plays a key role in immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

Regulation and Function:

  • AT-hook transcription factor AKNA regulates CD40 expression, important for homotypic cell interactions.
  • Adaptor protein TNFR2 interacts with CD40 to mediate signal transduction.
  • CD40-CD154 interaction is necessary for amyloid-beta-induced microglial activation, linked to Alzheimer’s disease pathogenesis.
  • Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
